چکیده

The aberrant differentiation of osteoclasts is a key feature the pathogenesis osteoporosis, which has devastating impact on human health. While effects Orientin (Ori) particularly RANKL-stimulated osteoclast production and activation, remain still unclear, Ori been found to display several biological activities, including antioxidant anti-inflammatory. In this work, we investigated possible pathways through suppressed RANKL-induced development showed for first time that it does so. macrophages from bone marrow (BMMs) were cultivated then treated with after being stimulated RANKL. Then, TRAP-positive multinucleated cells counted, F-actin ring analysis was used assess Ori's mature development. addition, dihydroethidium (DHE) staining evaluate reactive oxygen species (ROS). performed western blotting quantitative RT-PCR investigate probable causes these downregulation effects. We discovered inhibits creation osteoclasts, gene protein expressions unique ROS production. By activating Nrf2 other ROS-scavenging enzymes, reduces intracellular levels. expression main transcription factor development, c-Fos, downregulated together NFATc1, CTSK, NFATc2, thanks inhibition NF-κB. Consistent its in vitro antiosteoclastogenic action, therapy ovariectomized (OVX) rat model also able restore mass improve microarchitecture distal femurs. Together, our results demonstrate flavonoid molecule therapeutic promise illnesses associated such as osteoporosis.

Pu-erh Tea Extract Ameliorates Ovariectomy-Induced Osteoporosis in Rats and Suppresses Osteoclastogenesis In Vitro

Background and Objective: Tea drinking is associated with positive effects on bone health and may protect against osteoporosis, especially in elderly women. Pu-erh tea has many beneficial effects on human health; however, whether Pu-erh tea has anti-osteoporotic potential remains unclear. Thus, we investigated the effects of Pu-erh tea extract (PTE) on ovariectomy-induced osteoporosis in rats a...

Acteoside Suppresses RANKL-Mediated Osteoclastogenesis by Inhibiting c-Fos Induction and NF-κB Pathway and Attenuating ROS Production

Numerous studies have reported that inflammatory cytokines are important mediators for osteoclastogenesis, thereby causing excessive bone resorption and osteoporosis. Acteoside, the main active compound of Rehmannia glutinosa, which is used widely in traditional Oriental medicine, has anti-inflammatory and antioxidant potentials. In this study, we found that acteoside markedly inhibited osteocl...

Curcumin Ameliorates Furazolidone Induced DNA Damage and Apoptosis in Human Hepatocyte L02 Cells via Inhibiting ROS Production and Mitochondrial Pathway

Furazolidone (FZD), a synthetic nitrofuran derivative, has been widely used as an antibacterial and antiprotozoal agent. Recently, the potential toxicity of FZD has raised concerns, but its mechanism is still unclear. This study aimed to investigate the protective effect of curcumin on FZD induced cytotoxicity and the underlying mechanism in human hepatocyte L02 cells. The results showed that c...

Brazilin Ameliorates High Glucose-Induced Vascular Inflammation via Inhibiting ROS and CAMs Production in Human Umbilical Vein Endothelial Cells

Vascular inflammatory process has been suggested to play a key role in the initiation and progression of atherosclerosis, a major complication of diabetes mellitus. Recent studies have shown that brazilin exhibits antihepatotoxic, antiplatelet, cancer preventive, or anti-inflammatory properties. Thus, we investigated whether brazilin suppresses vascular inflammatory process induced by high gluc...

Vps35 loss promotes hyperresorptive osteoclastogenesis and osteoporosis via sustained RANKL signaling

Receptor activator of NF-κB (RANK) plays a critical role in osteoclastogenesis, an essential process for the initiation of bone remodeling to maintain healthy bone mass and structure. Although the signaling and function of RANK have been investigated extensively, much less is known about the negative regulatory mechanisms of its signaling. We demonstrate in this paper that RANK trafficking, sig...
